YEP broth has been used to culture Agrobacterium as well as yeasts(Schizosaccharomyces pombe and Saccharomyces cerevisiae.). YEP Broth is YPD (YEPD) without dextrose and is used as a base for making a variation of YPD with an alternate carbon source.

- Prepare 0.8 L of H2O in a suitable container.
- Add 20 g of Bacto peptone to the solution.
- Add 10 g of yeast extract to the solution.
- Add H2O until the volume is 1 L.
- Adjust pH to 7.0, and bring final volume to 1 L with H2O. Autoclave for 30 min on liquid cycle.
