Description
Substance P (SP) is a key neuropeptide that mediates diverse physiological actions by binding to the seven-transmembrane domain neurokinin 1 receptor (NK1-R). Upon binding to NK1-R on the cell surface, SP triggers clathrin-dependent endocytosis, with the ligand-receptor complex internalized into early endosomes containing transferrin receptors (Tf-R). In acidified perinuclear vesicles, SP dissociates from NK1-R—while SP is slowly degraded intracellularly, the NK1-R recycles back to the plasma membrane, a process independent of new protein synthesis but requiring endosomal acidification. This endocytic and recycling pathway contributes to the desensitization and resensitization of cellular responses to SP, highlighting SP’s critical role in regulating receptor-mediated signaling and cellular adaptation.
