Background
ADI-65534 is an engineered antibody variant derived from the pan-hantavirus protective antibody ADI-42898. It was designed to enhance the potency and cross-clade neutralization of the original antibody, particularly against the highly lethal Andes virus (ANDV). ADI-65534 was developed through affinity maturation of ADI-42898, resulting in improved binding affinity and reduced dissociation at acidic pH, which is crucial for its activity against ANDV. It recognizes a quaternary epitope spanning the Gn and Gc subunits of the hantavirus glycoprotein complex, locking the Gn/Gc heterodimer in its prefusion conformation and preventing membrane fusion, a critical step in viral entry. ADI-65534 maintains the broad-spectrum activity of its parent antibody, ADI-42898, and is effective against both Old World and New World hantaviruses.
