Super Broth is a robust L-Broth variant, nutritionally rich medium developed for growth and maintenance of recombinant E. coli strains and generates higher plasmid densities.

- Prepare 0.8 L of H2O in a suitable container.
- Add 32 g of tryptone to the solution.
- Add 20 g of yeast extract to the solution.
- Add 5 g of Sodium chloride to the solution.
- Add 0.2 g of Sodium Hydroxide to the solution.
- Add H2O until the volume is 1 L.
- Sterilize by autoclaving for 20 minutes on liquid cycle.
