SSC for Northerns (20×) Preparation and Recipe

SSC (saline sodium citrate) buffer is commonly used in Northern blotting as a hybridization or wash buffer.

To prepare L  SSC for Northerns (20×)

  1. Prepare 0.8 L of distilled water in a suitable container.
  2. Add 175.32 g of Sodium chloride to the solution.
  3. Add 77.42 g of Sodium citrate to the solution.
  4. Adjust the pH to 7.0.
  5. Add distilled water until the volume is 1 L.
  6. Autoclave the solution and store it at room temperature. It will last for 1 year or longer.
