SSC (saline sodium citrate) buffer is commonly used in Northern blotting as a hybridization or wash buffer.

- Prepare 0.8 L of distilled water in a suitable container.
- Add 175.32 g of Sodium chloride to the solution.
- Add 77.42 g of Sodium citrate to the solution.
- Adjust the pH to 7.0.
- Add distilled water until the volume is 1 L.
- Autoclave the solution and store it at room temperature. It will last for 1 year or longer.
