- Prepare 0.8 L of distilled water in a suitable container.
- Add 10 g of tryptone to the solution.
- Add 5 g of yeast extract to the solution.
- Add 5 g of Sodium chloride to the solution.
- Add 0.04 g of Sodium Hydroxide to the solution.
- Add distilled water until the volume is 1 L.
- Autoclave for 15 min on liquid cycle.
- Although the pH is adjusted to near 7 with NaOH, the medium is not very highly buffered, and the pH of a culture growing in the medium drops as the culture nears saturation.
