Description
SlRGF9 is a tyrosine-sulfated small secreted peptide belonging to the RGF/GLV peptide family in tomato. Its coding gene is specifically transcribed in tapetal cells and microspore mother cells of anthers upon low-temperature induction during reproductive development. Functionally redundant with SlRGF10, it acts as an extracellular ligand after post-translational tyrosine sulfation; mature sulfated SlRGF9 binds the plasma membrane receptor complex SlRGFR6-SlSERK to trigger downstream calcium influx via SlCNGC16/18 channels, which corrects delayed programmed cell death (PCD) of cold-stressed tapetum and safeguards normal pollen maturation under chilling conditions. Single knockout of SlRGF9 barely impairs cold tolerance because endogenous SlRGF10 compensates its physiological function, while simultaneous deletion of both SlRGF9 and SlRGF10 leads to severe pollen abortion and dramatic yield loss under low temperature. Exogenous application of purified sulfated SlRGF9 alone fully rescues the cold-sensitive phenotype of the double-knockout mutant. Non-sulfated SlRGF9 lacks receptor-binding affinity and shows no anti-cold bioactivity.
