    Ranacyclin-B-LK2 is a small serine protease inhibitor peptide identified from the skin secretion of the Ranidae frog species Limnonectes kuhlii. It belongs to the Ranacyclin-B peptide family, which is characterized by a conserved GCWTKSXXPKPC motif in the primary structure. The peptide is composed of 19–20 amino acid residues and is cationic, containing multiple basic residues (arginine and/or lysine) with no acidic amino acids. Ranacyclin-B-LK2 exhibits both trypsin-inhibitory and antimicrobial activities, functioning as a bi-functional defensive molecule in amphibian skin. The peptide adopts a conformation stabilized by a disulfide bond between cysteine residues at positions P4 and P7′, forming an 11-residue reactive loop (CWTKSPPKPC) that differs from the typical 9-residue loop found in other Bowman–Birk inhibitors. The trypsin-inhibitory constant (Ki) for Ranacyclin-B-LK2 is in the order of 10−6 M, and it demonstrates antimicrobial activity against Staphylococcus aureus with a minimal inhibitory concentration (MIC) of 11.8 μM.
