The phosphatase inhibitor pentapeptide 2 (NH2-ERGMT-COOH) is a specific pentapeptide inhibitor of Rap phosphatases in Bacillus subtilis. It is generated from the carboxy-terminal end of a precursor protein encoded by phr genes. The active pentapeptide form inhibits the phosphatase activity of Rap proteins, which dephosphorylate the Spo0F~P intermediate response regulator in the sporulation phosphorelay. This inhibition is part of a complex export-import processing pathway that regulates the production of the Phr pentapeptides. The structural organization of the Phr precursor suggests a SecA-dependent secretion pathway, but processing to the active pentapeptide occurs through a single cleavage event at the peptide bond on the amino-terminal end of the pentapeptide. This cleavage is independent of type I signal peptidase activity. The pentapeptide sequence is required to be at the carboxy-terminal end of the precursor, and an alanine residue at the -1 position preceding the pentapeptide is essential for correct processing.
