Description
The peptide PFVYLI is a short sequence derived from the C-terminal portion of the peptide C105Y (CSIPPEVKFNKPFVYLI). It is composed of six amino acids. PFVYLI was investigated to determine the sequence responsible for the internalization and nuclear localization of the C105Y peptide. PFVYLI alone could mediate cellular uptake and nuclear targeting of C105Y, as it was internalized by HuH7 cells and displayed a staining pattern identical to C105Y. Additionally, the peptide lacking the PFVYLI sequence (FVYLI) was not taken up by HuH7 cells, indicating the importance of this sequence for the uptake of the peptide. Furthermore, the order of the amino acids within the PFVYLI sequence did not appear to influence cell penetration and nuclear localization, as the scrambled mutant (SCRAM) with the same amino acids in a different order was also internalized by the cells with a similar staining pattern. PFVYLI is a key sequence within the C105Y peptide that plays a crucial role in its cellular uptake and nuclear targeting.

About TFA salt
Trifluoroacetic acid (TFA) has a significant impact on peptides due to its role in the peptide synthesis process.
TFA is essential for the protonation of peptides that lack basic amino acids such as Arginine (Arg), Histidine (His), and Lysine (Lys), or ones that have blocked N-termini. As a result, peptides often contain TFA salts in the final product.
TFA residues, when present in custom peptides, can cause unpredictable fluctuations in experimental data. At a nanomolar (nM) level, TFA can influence cell experiments, hindering cell growth at low concentrations (as low as 10 nM) and promoting it at higher doses (0.5–7.0 mM). It can also serve as an allosteric regulator on the GlyR of glycine receptors, thereby increasing receptor activity at lower glycine concentrations.
In an in vivo setting, TFA can trifluoroacetylate amino groups in proteins and phospholipids, inducing potentially unwanted antibody responses. Moreover, TFA can impact structure studies as it affects spectrum absorption.
