2',5'-phosphodiesterase 12 (PDE12) is a mitochondrial enzyme that plays a critical role as a negative regulator of the 2-5A system, a major pathway for antiviral and antitumor functions induced by interferons. The enzyme cleaves both 2',5'-phosphodiester bonds in triphosphorylated oligoadenylates (2-5A) and 3',5'-phosphodiester bonds of oligoadenylates, degrading 2-5A to produce AMP and ATP. By suppressing cellular 2-5A levels, PDE12 modulates antiviral responses, and its inhibition has been shown to decrease viral replication in cultured cells. The protein localizes to the mitochondrial matrix, where it performs its regulatory functions in innate immunity pathways.
