    Neuropeptide S (NPS) is a conserved neuropeptide that activates its cognate G protein-coupled receptor (NPSR), leading to mobilization of intracellular calcium. The precursor mRNA for NPS is expressed in a discrete cluster of neurons located between the locus coeruleus and Barrington's nucleus in the brainstem. The receptor for NPS is widely distributed in the brain, with high expression levels in regions such as the amygdala, thalamic midline nuclei, and cortical areas.

    Central administration of NPS modulates behavioral states, increasing locomotor activity and wakefulness while reducing slow-wave and rapid-eye-movement sleep in rodents. The anatomical distribution of the NPS system and its functional effects suggest it is a modulator of arousal and related behaviors.
