  • Description

    Neuropeptide S (NPS) is a neuropeptide that potently modulates arousal and exhibits anxiolytic-like behavioral effects. It acts by activating its cognate G protein-coupled receptor (NPSR), which is widely distributed in the brain, including regions such as the amygdala and thalamic nuclei. The precursor mRNA for NPS is expressed in a discrete cluster of cells located between the locus coeruleus and Barrington's nucleus in the brainstem, defining a previously uncharacterized neuronal population.

    Central administration of NPS increases locomotor activity and promotes wakefulness while reducing slow-wave and REM sleep in rodents. The peptide also produces anxiolytic-like effects in multiple behavioral paradigms, such as the elevated plus maze and light-dark box, independent of its arousal-promoting properties. These findings indicate that NPS is a novel modulator involved in regulating states of arousal and anxiety-related behaviors.
