Description
FPGERGVEGPGP polypeptide is an eight-amino acid sequence within the alternatively spliced FN - III repeat D of human tenascin - C. It has been identified as crucial for neurite outgrowth promotion. This peptide is active by itself and also functions in the context of the large tenascin - C splice variant. The biological activity of FPGERGVEGPGP depends on specific amino acids within the sequence.
It can overcome the inhibitory effect of chondroitin sulfate proteoglycans (CSPGs) on neurite outgrowth in vitro, which is significant as CSPGs are upregulated in glial scars and impede axonal regrowth. In neural tissue engineering applications, it has the potential to enhance neuronal regeneration and differentiation. For example, in studies with PC12 cells, related peptide nanofibers have been shown to increase the length and number of neurites in 2D and 3D cultures, and promote the expression of neural markers such as β - III tubulin and synaptophysin, suggesting its possible role in the development of therapies for neurological conditions. -

About TFA salt
Trifluoroacetic acid (TFA) has a significant impact on peptides due to its role in the peptide synthesis process.
TFA is essential for the protonation of peptides that lack basic amino acids such as Arginine (Arg), Histidine (His), and Lysine (Lys), or ones that have blocked N-termini. As a result, peptides often contain TFA salts in the final product.
TFA residues, when present in custom peptides, can cause unpredictable fluctuations in experimental data. At a nanomolar (nM) level, TFA can influence cell experiments, hindering cell growth at low concentrations (as low as 10 nM) and promoting it at higher doses (0.5–7.0 mM). It can also serve as an allosteric regulator on the GlyR of glycine receptors, thereby increasing receptor activity at lower glycine concentrations.
In an in vivo setting, TFA can trifluoroacetylate amino groups in proteins and phospholipids, inducing potentially unwanted antibody responses. Moreover, TFA can impact structure studies as it affects spectrum absorption.
