    Melanin-concentrating hormone (MCH) is a cyclic neuropeptide, 19 amino acids in length, predominantly synthesized by neurons in the hypothalamus. The PMCH gene encodes a precursor protein, PMCH, from which mature MCH is cleaved at dibasic proteolytic sites. This gene is conserved across a wide range of vertebrates, with its structure comprising three exons and two introns in mammals, teleosts, and other groups, although a teleost-specific, intronless paralog (pmchb) also exists. The peptide is characterized by a disulfide bridge between cysteine residues 7 and 16, forming a ring structure essential for receptor binding. The MCH system exhibits significant morphological diversity across species. In early chordates like lampreys, MCH neurons are found in the periventricular dorsomedial hypothalamic nucleus, often contacting the third ventricle. In ray-finned fishes, neurons are mainly located in the nucleus lateralis tuberis and project prominently to the pituitary. In mammals, the predominant MCH neuronal population migrates laterally to the lateral hypothalamic area, with a smaller group in the dorsomedial hypothalamus and zona incerta, establishing widespread central nervous system projections. This evolutionary trajectory reflects a shift from periventricular and neuroendocrine roles in ancestral vertebrates to a broader integrative function in the mammalian brain, where the peptide modulates arousal, motivated behaviors, and energy balance through its receptor, MCHR1 (and MCHR2 in most non-Glires mammals).
