    Dynorphin B [1-29] was found in pig (Sus scrofa). Porcine dynorphin B-29, also referred to as leumorphin or prodynorphin (228-256), is a 29-amino acid endogenous opioid peptide derived from the precursor proenkephalin B. Its sequence is Tyr-Gly-Gly-Phe-Leu-Arg-Arg-Gln-Phe-Lys-Val-Val-Thr-Arg-Ser-Gln-Glu-Asp-Pro-Asn-Ala-Tyr-Tyr-Glu-Glu-Leu-Phe-Asp-Val. This peptide was chemically synthesized using solid-phase methods and purified through gel filtration, ion exchange, reverse phase liquid chromatography, and preparative high pressure liquid chromatography, achieving a purity of at least 99% as confirmed by HPLC, thin-layer chromatography, and paper electrophoresis. In binding experiments using mouse brain tissue, dynorphin B-29 displays equal affinity for μ and κ opioid receptors (KI values of 5.0 ± 0.7 nM and 5.3 ± 0.58 nM, respectively) and significant but lower affinity for δ receptors (KI of 18.4 ± 2.4 nM). In isolated tissue preparations, its potency to inhibit electrically evoked contractions varies across systems, being highest in the guinea pig ileum (IC50 of 7.54 ± 1.8 nM), followed by mouse vas deferens, rabbit vas deferens, and rat vas deferens, where only partial inhibition is observed. The presence of peptidase inhibitors further reduces IC50 values, particularly in rabbit vas deferens, indicating that the peptide is subject to enzymatic degradation in different tissues.
