Description
Klotho-derived peptide 1 hydrochloride (KP1) is a 30-amino acid peptide corresponding to Phe57–Lys86 of human Klotho, representing a buried β-strand in the N-terminal domain. Chemically synthesizable and cost-effective, KP1 recapitulates Klotho's anti-fibrotic and renoprotective actions. KP1 binds TGF-β receptor 2 (TβR2), disrupting TGF-β/TβR2 interaction and inhibiting downstream Smad2/3 and MAPK pathways. In mouse models of renal fibrosis, KP1 preserves kidney function, ameliorates fibrosis, and restores endogenous Klotho expression via posttranscriptional regulation through the miR-223-3p/lncRNA-TUG1 axis. Additionally, KP1 protects against acute kidney injury by directly targeting mitochondrial ATAD3A, preventing cytochrome c release and tubular apoptosis—independent of TGF-β signaling. KP1 demonstrates efficacy across multiple renal injury models (UUO, UIRI, cisplatin, IRI).
