    HKPao, also known as Adipotide, is a synthetic hunter-killer peptide composed of two distinct domains. The amino-terminal homing sequence specifically interacts with cell surface receptors, facilitating targeted intracellular delivery. This domain is connected via a glycinyl glycine linker to a conserved carboxyl-terminal pro-apoptotic sequence, which consists of D-amino acids to enhance stability.

    The peptide enters cells through receptor-mediated interactions and migrates toward mitochondria. Experimental studies demonstrate that HKPao induces the formation of transient pores in phospholipid vesicle membranes without causing lysis. These pores are sufficiently large to permit the efflux of entrapped cytochrome c, indicating a membrane-perturbing mechanism. The peptide exhibits a graded mode of leakage, where vesicles retain partial contents post-treatment, and the pore size allows selective release of molecules based on hydrodynamic radius.
