    The HER2/neu-derived peptide GP2, with the amino acid sequence IISAVVGIL, corresponds to residues 654-662 of the HER2/neu protein. This nonapeptide is an HLA-A2-restricted tumor antigen capable of inducing cytotoxic T lymphocytes that react to various epithelial cancers. However, the native GP2 peptide exhibits very low binding affinity for the HLA-A2 molecule, primarily due to subdominant amino acids at its major anchor positions.

    Modifications involving amino acid substitutions, particularly at position 1, can significantly enhance GP2's binding affinity to HLA-A2. For instance, the modified peptide 1F2L10V (sequence FLSAVVGILV) demonstrates improved stabilization of the peptide-HLA-A2 complex and is effectively recognized by GP2-specific cytotoxic T lymphocytes. These alterations increase the immunogenic potential of the peptide while maintaining recognition by T cell receptors directed against the native epitope.
