Description
Hainanenin-1 is a 21-amino acid antimicrobial peptide originally purified from the skin secretion of the Hainan cascade-frog, Amolops hainanensis. It belongs to the hainanenin family, a novel group of amphibian antimicrobial peptides characterized by a C-terminal disulphide loop of seven residues formed between Cys15 and Cys21. The peptide contains three basic residues (two lysines and one arginine) with no acidic residues, resulting in a net charge of +3. Structural analysis indicates that hainanenin-1 adopts an amphipathic alpha-helical conformation, a feature common among many antimicrobial peptides and considered important for their biological function.Hainanenin-1 exhibits strong and broad-spectrum antimicrobial activity against Gram-positive bacteria, Gram-negative bacteria, and fungi, including several resistant strains. It also displays slight antioxidant activity in DPPH free radical scavenging assays, but shows relatively strong hemolytic activity on human erythrocytes. The discovery of hainanenin-1 increases the diversity of known amphibian antimicrobial peptides and provides new structural templates for anti-infective agent design.
