    The DDDEEKC peptide is a bioinspired sequence derived from the N-terminal region of statherin, a salivary acquired pellicle protein. This heptapeptide exhibits a high affinity for hydroxyapatite surfaces due to its anionic character, primarily conferred by the aspartic acid (D) and glutamic acid (E) residues. The adsorption capacity is attributed to the peptide's negative charge density and its potential to adopt specific conformations, such as an α-helical structure, which facilitate strong interactions with the calcium ions in the hydroxyapatite lattice.

    Functionally, the DDDEEKC sequence serves as an effective adhesion motif, enabling targeted binding to mineralized substrates like tooth enamel. When conjugated to other molecular entities, such as polyphenol-based compounds like tannic acid, the peptide directs the assembly and localization of the conjugate onto hydroxyapatite. This specific adsorption provides a stable interface for subsequent processes, such as the templated growth of hydroxyapatite crystals from calcium and phosphate ions, mimicking aspects of natural biomineralization.
