Description
CW49 is a small peptide composed of 11 amino acid residues with the sequence APFRMGICTTN, identified from the skin of the frog Odorrana grahami. This peptide exhibits notable pro-angiogenic activity, as demonstrated by its ability to induce tube formation in human umbilical vein endothelial cells in vitro, an effect comparable to that of vascular endothelial growth factor. In a murine full-thickness dermal wound model, CW49 enhances wound healing by promoting the formation of granulation tissue and modulating epidermal thickness. Mechanistically, it upregulates the expression of key angiogenic proteins, including hypoxia-inducible factor-1 alpha, endothelial nitric oxide synthase, and inducible nitric oxide synthase, without significantly altering vascular endothelial growth factor levels. In addition to its angiogenic properties, CW49 displays anti-inflammatory activity by reducing the infiltration of macrophages and decreasing the levels of pro-inflammatory cytokines such as interleukin-6 and tumor necrosis factor-alpha in wound tissue. In vitro experiments with lipopolysaccharide-stimulated macrophages confirm that CW49 inhibits the secretion of these cytokines while promoting inducible nitric oxide synthase expression and nitric oxide production. This peptide does not significantly affect re-epithelialization or wound closure rates, suggesting its primary roles involve angiogenesis and modulation of the inflammatory response. CW49 represents a promising candidate for further investigation as a bioactive molecule with potential applications in tissue repair and regenerative biology.
