Description
A novel 29-amino acid natriuretic peptide was identified in chicken heart tissue, purified by monitoring its relaxant effect on chick rectum. The peptide, designated chicken α-ANP (α-chANP), was isolated from both ventricular and atrial extracts and its complete amino acid sequence was determined. The sequence shows remarkable homology with mammalian atrial natriuretic peptide (ANP), particularly within the 17-amino acid ring structure formed by a disulfide linkage, which is essential for biological activity. However, maximum sequence homology was observed between chicken α-ANP and porcine brain natriuretic peptide (BNP), indicating a closer evolutionary relationship to BNP than to mammalian ANP. Unique structural features were noted at the N- and C-terminal regions, including a C-terminal Lys-Asn sequence that differs from the conserved Arg-Tyr found in both mammalian ANP and BNP.
