    TPGWamide is a tetrapeptide belonging to the APGWamide-related neuropeptide family, identified in the cephalopod mollusk Sepia officinalis. This peptide, with the sequence Thr-Pro-Gly-Trp-NH2, is a product of the Mytilus edulis gene and is processed by a dipeptidyl aminopeptidase (DPAP). Tissue mapping via LC-ESI-MS/MS analysis reveals that TPGWamide is exclusively localized within the central nervous system (CNS) of S. officinalis.

    In vitro processing studies demonstrate that TPGWamide can be cleaved by DPAP to yield the dipeptide GWamide, although its conversion efficiency is comparatively low. Among related tetrapeptides, TPGWamide shows the lowest processing rate, which is attributed to the influence of its N-terminal threonine residue on DPAP substrate specificity. The presence and specific CNS localization of TPGWamide suggest it is part of a neuropeptide family involved in diverse regulatory functions within molluskan nervous systems.
