Description
3×HA Peptide is a synthetic peptide presenting three tandem copies of the influenza hemagglutinin (HA) epitope (YPYDVPDYA), intended as a competitive eluent for anti-HA affinity matrices. It corresponds to three tandem copies of the hemagglutinin (HA) epitope derived from the human influenza virus HA protein. The triple arrangement of the epitope is designed to enhance affinity toward anti-HA antibodies compared to a single HA tag.
The 3×HA peptide is used as a competitive eluent in co-immunoprecipitation procedures. When 3×HA-tagged bait proteins are captured by anti-HA antibodies immobilized on magnetic beads or agarose resin, the addition of excess 3×HA peptide displaces the tagged bait from the antibody through competitive binding. This allows elution of 3×HA-tagged proteins and their associated complexes in native form, preserving protein structure and biological activity, while matrix-bound unspecific proteins and antibodies remain on the support. Optimal elution conditions, including peptide concentration, temperature, and incubation time, are typically determined empirically to maximize yield and purity.
