    1018M is a novel antimicrobial peptide designed through molecular docking and amino acid substitution based on the innate defense regulator peptide IDR-1018. It features an α-helix structure and exhibits enhanced physicochemical properties, including a higher positive charge and isoelectric point compared to its parental peptide. The peptide demonstrates strong binding affinity to the stringent response signaling molecule ppGpp, forming multiple hydrogen bonds through specific amino acid residues.

    This peptide effectively disrupts bacterial cell walls and permeabilizes cell membranes, leading to significant structural damage and content leakage. Additionally, 1018M interacts with bacterial genomic DNA and regulates the expression of genes related to ppGpp metabolism and biofilm formation. It displays improved stability under varying temperatures, pH conditions, and protease exposure, contributing to its robust antimicrobial and anti-biofilm activities.
